Determination of rational pH for carbomer-based dental gel by texture analysis

Introduction: The significant influence on the oral cavity tissues health has a pH value, that’s why when developing dental medicines, it is necessary to keep this indicator within the rational limits, which corresponds to the value of saliva pH (5.5-8.0) [1]. For carbomer-based gels the required pH can be created by neutralizing the gel, resulting in a change of its structure [2]. Therefore, the purpose of our work was to evaluate the effect of pH on the textural properties of the dental gel being developed. Materials and methods: The samples of combined composition dental gel on the basis of carbopol Polacril® 40P, neutralized with 10 % sodium hydroxide solution until pH values 5.54, 6.02 and 6.58 were used in the study. The textural properties (backward extrusion and spreadability tests) were studied using a TA.XT Plus Texture Analyzer (UK) [3]. The analysis was carried out in comparison with the drug “Dentinox-Gel N” (Dentinox Gesellschaft für Pharmaceuticals, Praparate Lenk & Schuppan, Germany), which are also carbomer based and has pH = 6.25. Results: Dental gels based on the carbopol Polacril® 40P revealed a linear dependence between pH values and textural properties. The tested gels proved good spreadability, cohesion and adhesion properties. It will allow to withstanding the physiological damage caused by jaw movement and saliva flow, and provide intimate and sustainable contact of the gel with oral tissues [4]. Conclusions: Study on the textural properties of a dental medicine revealed that the behavior of carbopol gels in the pH range 5.5-6.5 had not changing significantly but the most similar results to the comparison drug had a gel with a pH 6.02, that will ensure a long contact time of the drug in the site of application and its high bioavailability.
